Some compounds are badly or not at all visible for common detectors.
Post-column derivatization enhances sensitivity and selectivity for many HPLC applications. The trick: Right after the analytical column the analyte is changed "online" in a chemical reaction. The product then has physical and chemical properties (e.g. an elevated fluorescence), rendering it visible for the detector.
